    Cardiovascular responses to postural change and aerobic capacity among middle-aged men and women before and after aerobic physical training

    OBJETIVO: Comparar respostas cardiovasculares a Manobra Postural Passiva (Tilt Test) e capacidade cardiorrespiratória em homens e mulheres de meia-idade antes e após treinamento físico aeróbio. MATERIAIS E MÉTODOS: Sete homens - GH (44,6±2,1 anos) e sete mulheres - GM (51,7±4,8 anos), participaram de treinamento físico aeróbio por 12 semanas. Foi realizado protocolo de Tilt Test (cinco minutos supino, dez minutos inclinado 70º, cinco minutos supino), com monitoração da pressão arterial e freqüência cardíaca. Para mensuração da capacidade cardiorrespiratória foi realizado protocolo em cicloergômetro. RESULTADOS: Na condição sedentária, GH mostrou maior influência parassimpática no controle da freqüência cardíaca evidenciada por maior intervalo RR (iRR) durante Tilt Test. Na condição treinada, os valores de iRR de ambos os grupos se assemelham, tendo as mulheres iRR maior em supino, mas na inclinação os homens mantêm iRR mais elevado. Para pressão arterial, as mulheres permanecem com valores superiores após treino, mas a freqüência cardíaca tende a se assemelhar em ambos. Já na capacidade cardiorrespiratória, homens e mulheres têm um padrão de comportamento semelhante após treino. Com exceção dos valores absolutos da freqüência cardíaca, sem diferenças entre os grupos, para todas as outras variáveis os homens obtiveram valores superiores aos das mulheres. Observa-se ainda que, após o treinamento, houve redução significativa dos valores de pressão arterial no GM, mesmo continuando superiores aos dos homens. CONCLUSÃO: O treinamento parece ter reduzido os níveis pressóricos nas mulheres, além de serem observadas melhorias na capacidade cardiorrespiratória de ambos os grupos, permanecendo os homens com melhor desempenho do que as mulheres.OBJECTIVE: To compare the cardiovascular responses to passive postural maneuvers (tilt test) and the cardiorespiratory capacity in middle-aged men and women, before and after aerobic physical training. RESULTS: In the sedentary condition, men showed greater parasympathetic influence in heart rate control, as demonstrated by their higher RR interval (iRR) during the tilt test. After training, the iRR values became more similar in the two groups, although the women had higher iRR in the supine position and the men continued to present higher iRR under tilted conditions. The women's blood pressures continued to be higher after training, but heart rate tended to become similar in the two groups. The cardiorespiratory capacity patterns in the two groups were similar after training. Except for absolute heart rate values, for which there were no differences between the groups, the men's values were higher than those of the women for all other variables. It was also observed that, after the training, the women's blood pressures were significantly lower, even though their pressures remained higher than the men's. CONCLUSIONS: The training seemed to reduce the women's arterial blood pressure levels and improve both groups' cardiorespiratory capacity, but the men continued to present better performance than the women

    Characterization of mycobacteria and mycobacteriophages isolated from compost at the São Paulo Zoo Park Foundation in Brazil and creation of the new mycobacteriophage Cluster U

    Background: A large collection of sequenced mycobacteriophages capable of infecting a single host strain of Mycobacterium smegmatis shows considerable genomic diversity with dozens of distinctive types (clusters) and extensive variation within those sharing evident nucleotide sequence similarity. Here we profiled the mycobacterial components of a large composting system at the São Paulo zoo. Results: We isolated and sequenced eight mycobacteriophages using Mycobacterium smegmatis mc2155 as a host. None of these eight phages infected any of mycobacterial strains isolated from the same materials. The phage isolates span considerable genomic diversity, including two phages (Barriga, Nhonho) related to Subcluster A1 phages, two Cluster B phages (Pops, Subcluster B1; Godines, Subcluster B2), three Subcluster F1 phages (Florinda, Girafales, and Quico), and Madruga, a relative of phage Patience with which it constitutes the new Cluster U. Interestingly, the two Subcluster A1 phages and the three Subcluster F1 phages have genomic relationships indicating relatively recent evolution within a geographically isolated niche in the composting system. Conclusions: We predict that composting systems such as those used to obtain these mycobacteriophages will be a rich source for the isolation of additional phages that will expand our view of bacteriophage diversity and evolution

    Oral tolerance to cancer can be abrogated by T regulatory cell inhibition

    Oral administration of tumour cells induces an immune hypo-responsiveness known as oral tolerance. We have previously shown that oral tolerance to a cancer is tumour antigen specific, non-cross-reactive and confers a tumour growth advantage. We investigated the utilisation of regulatory T cell (Treg) depletion on oral tolerance to a cancer and its ability to control tumour growth. Balb/C mice were gavage fed homogenised tumour tissue – JBS fibrosarcoma (to induce oral tolerance to a cancer), or PBS as control. Growth of subcutaneous JBS tumours were measured; splenic tissue excised and flow cytometry used to quantify and compare systemic Tregs and T effector (Teff) cell populations. Prior to and/or following tumour feeding, mice were intraperitoneally administered anti-CD25, to inactivate systemic Tregs, or given isotype antibody as a control. Mice which were orally tolerised prior to subcutaneous tumour induction, displayed significantly higher systemic Treg levels (14% vs 6%) and faster tumour growth rates than controls (p<0.05). Complete regression of tumours were only seen after Treg inactivation and occurred in all groups - this was not inhibited by tumour feeding. The cure rates for Treg inactivation were 60% during tolerisation, 75% during tumour growth and 100% during inactivation for both tolerisation and tumour growth. Depletion of Tregs gave rise to an increased number of Teff cells. Treg depletion post-tolerisation and post-tumour induction led to the complete regression of all tumours on tumour bearing mice. Oral administration of tumour tissue, confers a tumour growth advantage and is accompanied by an increase in systemic Treg levels. The administration of anti-CD25 Ab decreased Treg numbers and caused an increase in Teffs. Most notably Treg cell inhibition overcame established oral tolerance with consequent tumor regression, especially relevant to foregut cancers where oral tolerance is likely to be induced by the shedding of tumour tissue into the gut

    Effects of chronic exercise on severity, quality of life and functionality in an elderly Parkinson’s disease patient: case report

    Exercise produces potential influences on physical and mental capacity in patients with neuropsychiatric disor- ders, and can be made a viable form of therapy to treat Parkinson’s disease (PD). We report the chronic effects of a regu- lar physical exercise protocol on cognitive and motor functions, functional capacity, and symptoms in an elderly PD pa- tient without dementia. The patient participated of a program composed of proprioceptive, aerobic and flexibility exer- cises, during 1 hour, three days a week, for nine months. Patient used 600 mg of L-DOPA daily, and 1 hour prior to each exercise session. Assessment was conducted in three stages, 0-3, 3-6 and 6 to 9 months, using percentual variation to the scales Hoehn and Yahr, Mini-Mental State Examination (MMSE), Parkinson Activity Scale (PAS), Beck Depression In- ventory (BDI), and Unified Parkinson's Disease Rating Scale (UPDRS-III). Reassessment showed clear changes in clini- cal parameters for Hoehn and Yahr (4 to 2.5), MMSE (14 to 22), PAS (13 to 29), BDI (9 to 7) and UPDRS-III (39 to 27) at the end of 9 months. According to our data, exercise seems to be effective in promoting the functional capacity and the maintenance of cognitive and motor functions of PD patients. Regular exercise protocols can be implemented as an ad- junctive treatment for reducing the severity of PD

    Synthesis and Anti-Mycobacterium tuberculosis Activity of Imidazo[2,1-b][1,3]oxazine Derivatives against Multidrug-Resistant Strains

    The emergence of multidrug-resistant strains of M. tuberculosis has raised concerns due to the greater difficulties in patient treatment and higher mortality rates. Herein, we revisited the 2-nitro-6,7-dihydro-5H-imidazo[2,1-b][1,3]oxazine scaffold and identified potent new carbamate derivatives having MIC90 values of 0.18–1.63 μM against Mtb H37Rv. Compounds 47–49, 51–53, and 55 exhibited remarkable activity against a panel of clinical isolates, displaying MIC90 values below 0.5 μM. In Mtb-infected macrophages, several compounds demonstrated a 1-log greater reduction in mycobacterial burden than rifampicin and pretomanid. The compounds tested did not exhibit significant cytotoxicity against three cell lines or any toxicity to Galleria mellonella. Furthermore, the imidazo[2,1-b][1,3]oxazine derivatives did not show substantial activity against other bacteria or fungi. Finally, molecular docking studies revealed that the new compounds could interact with the deazaflavin-dependent nitroreductase (Ddn) in a similar manner to pretomanid. Collectively, our findings highlight the chemical universe of imidazo[2,1-b][1,3]oxazines and their promising potential against MDR-TB

    Longitudinal differentiation in Melipona mandacaia (Hymenoptera, Meliponini) chromosomes

    Melipona manducaia is a stingless bee endemic to northeast Brasil. We describe the M. mandacaia karyotype using C-banding technique, fluorochrome staining and treatment with restriction enzymes and discuss the position of this species in the context of the phylogeny of the genus. Melipona monducaia has 2n = 18 (14 SM + 2 M + 2 A). Heterochromatin was detected in the pericentromeric region of pairs 1, 2 and 8 and in the form of small blocks in the remaining pairs. Staining with base-specific fluorochromes showed that this heterochromatin was rich AT (QM and DAPI), except in the region corresponding to the NOR which was rich GC (CMA(3)) and was cleaved by the HaeIII enzyme. Melipona manducaia is a member of Group I Melipona. Treatment with DraI/Giemsa discloses a larger number of bands than treatment with DraI/QM. Pre-cleavage with DraI gave rise to a larger number of bands following QM staining; a circumstance evidently due to a removal of the DNA-protein complex that prevented the association of the fluorochrome with AT-rich DNA. The results highlight the complex nature of heterochromatin.138213313

    Keratocystic odontogenic tumor overexpresses invadopodia-related proteins, suggesting invadopodia formation

    OBJECTIVE: Keratocystic odontogenic tumor (KOT) is an odontogenic neoplasm that shows aggressive clinical behavior and local invasiveness. Invadopodia are actin-rich cellular protrusions exhibiting proteolytic pericellular activity, thereby inducing focal invasion in neoplastic cells and increasing neoplasms aggressiveness. Thus, this study aimed to evaluate immunoexpression of invadopodia-related proteins, cortactin, MT1-MMP, Tks4, and Tks5, in KOT. STUDY DESIGN: Immunohistochemistry of 16 cases of KOT, eight cases of calcifying cystic odontogenic tumor (CCOT), and eight samples of the oral mucosa (OM) was carried out to assess the expression of the above described invadopodia-related proteins in the basal and suprabasal layer. RESULTS: KOT samples showed higher and significant immunoexpression of cortactin, MT1-MMP, TKs4, and TKs5 compared with the CCOT and OM samples. Significant expression of all these proteins was observed in the basal layer compared with the suprabasal layer in KOT. CONCLUSIONS: Overexpression of cortactin, MT1-MMP, TKs4, and TKs5 was observed in KOT compared with samples of CCOT and OM. These proteins were also overexpressed in the basal over the suprabasal layer of KOT samples. Taken together, these results suggest the participation of invadopodia-related proteins on the pathogenesis of this lesion
